Pure Gym has set its sights on the former Fitness First base in Level Street, which closed last year.
The new 25,402 sq ft gym will employ between 12 and 15 staff and will be open 24 hours a day.
Pure Gym spokesman Thomas Inskip said the centre was expected to open in May. Work to re-fit the building is currently being carried out.
The launch will follow the opening of branches in West Bromwich and Wolverhampton in May and in Walsall in June last year.
The new base in Brierley Hill is part of a £37 million expansion project. Pure Gym started in 2009 with four clubs.
It now has 43 gyms open across the UK.
